Methodology: DNA extraction and amplification by real-time polimerase chain reaction using primers specific for the region containing the JAK2 gene analyzed residue. Detection is performed by hybridization with FRET probes and melting curve analysis. Analytical sensitivity and specificity are 1% and >95% respectively. NOTE: More than 90% of patients with Polycythemia Vera and 50% of patients with Essential Thrombocythemia or Primary Myelofibrosis carry the V617F somatic mutation in the JAK2 gene (Nature. 2005 Apr 28;434(7037):1144-8, N Engl J Med. 2005 Apr 28;352(17):1779-90).
